It has been reported that motilin and ghrelin induce phase III-like contraction of the migrating motor complex (MMC). However, the detailed mechanisms by which motilin and ghrelin induce contraction of the MMC are still unclear because of the lack of small animal models such as rats and mice. To obtain a clearer insight into the mechanisms, we focused on the Suncus murinus as a small model animal for motilin and ghrelin study on the gastric contraction in vivo. A strain gauge transducer was sutured to the stomach and gastric contraction activity was monitored using conscious suncus. During the fasted state, clear phase I, phase II and phase III contractions were observed in the gastric body as found in humans and dogs, and typical phase III contractions at 80-150min intervals were also found. Bolus injection of motilin induced gastric phase III-like contraction in a dose-dependent manner. Intravenous infusion of ghrelin did not induce gastric contraction in the first half of phase I but significantly evoked gastric contraction in the latter half of phase I. In addition, ghrelin administration in phase II also induced gastric contraction in a dose-dependent manner. The results strongly suggest that these family peptides are involved in the interdigestive MMC in the suncus stomach. |
