IF4b-1 Viral Factors in HBV-associated Hepatocarcinogenesis

抄録 More than 30 thousand individuals die of hepatocellular carci’noma (HCC) in Japan in recent years. Amongthosehepatitis C virus (HCV)一associa/ted HCC accounts for nearly 700/obeing the leading cause of HCCsince the identification of HCV in 1989. Hepatitis B virus (HBV)which is endemic in most Asian countriesaccounts for only 10一一15P/o of HCC death in Japan. Nonethelessthe number of HCC death related to HBVinfection has been stable over these 30 yearssti11 an important cause of HCC in Japan.Among numerous factors supposed as the risk factor$ for hepatocarcinogenesis in chronic HBV infectionsuch as inflamm.ation or aflatoxinthe且BV itself has also been demonstrated to be responsible for HCCdevelopment as the viral factorboth in clinical and experimental studies. ln experimental studiesthe in-tegration of HBV genome into the HCC genome was discovered in 1980’sand the integration site analyseshad been performed meticulously. Howeverthe accumulated data resulted in a wide variation of HBV in-tegration sitesfailing to identify a spechic mechani$m that the activation of oncogenes or inactivation oftumor suppressor genes is caused by HBV. Alternativelya viral gene productHBxwhich works as atranscriptional factor in HBV replicationhas been shown to be involved in hepatocarcinogenesisusingboth cu.ltured cells and mouse models. Subsequentlythe contribution of HBx has been evidenced in且BV-associated HCC also using human HCC samples. Such a story of searching the mechanism of HBV-associated hepatocarcinogenesis will be presented.
