Chronic HBV infection has been recognized as one of iMportant cause for liver cancerespecially in theAsia. lt is generally accepted that the carcingogenesis develops via long-term chronic infections and sub-sequent accumulation of genetic mutationswhich finally transform the infected hepatocytes. The presen-tation will discuss the importance of viral factors at first. Epidemiological studies supported a high viralloadcertain viral genetic variants carrying higher risks for HCC.Regarding the host factors influencing liver carcinogenesisthese include the old-agesfamily history andgenderThe male gender is more susceptible to HBV-related H:CC than females. Our studies in ce田ineand in mouse model discovered an promoting role of androgen pathways and in contrastan suppressingrole of estrogen pathway in HBV-related HCCs. This may partially explained the male susceptibdity toHBV-related HCC. ln search for heredity in HBV-related HCCswe previously showed an increased riskamong H BV carriers with familial history of HCC. Now we initiated a genome-wide association study tosearch for possible susceptibility genetic loci. The stage 1 GWAS (Genome-Wide-Association Study) wascompleted and c andidate SNPS are under stage II scrutinizing. The results may shed lights on the mecha-nisms of HBV-related HCCs. There are recently several signaling pathwaysincluding RasWntAkt mole-culesto be involved in maintaining HCC transformation phenotypes. Current molecular therapies target-ing to these pathways may bring new hopes for HCC treatment |
