IF-BP2-2 Colitogenic Effector Memory CD4+ T Cells Develop Th17 Mediated Interstitial Pneumonia Independent to Intestinal Bacterial Antigens

抄録 Background : Although inflammatory bowel diseases (IBD) often include various extraintestinal manifes-tationslittle is known about their pathogenesis. Recentlywe have reported thatcolitogenic memoryCD4’ T cells were retained outside the intestine independent to intestinal bacterial antigens.’Aim and Methods : To clarify the role of colitogenic memory CD 4’ T cells for the pathogenesis of ex-traintestinal manifestations of IBDwe have investigated extraintestinal lesion pathologically throughoutthe body of colitis mice.Results:Colitic SCID mice previously transferred with CD4+CD45RBhigh T ce皿s developed hyperplasia ofinducible bronchus associated lymphoid tissues (iBALT)/ and interstitial pneumoniawhich was sup-pressed completely by transferring with C.D4+CD25+regulatory T cells. On the other handora1 admini-stration of antibiotics(ABX)or Ger血free(GF)condition coUld not prevent these lung manifestationswhile they completely prevented to develop colitis. A large number of CD4’CD44’CD62L’IL-7Rcthigh’ effec-tor memory T cells (TEM) infiltrate d in the iBALT and interstitial tissue of lung from colitic SCID mice.These lung CD4’ TEM produced large amount of IL-17 with the stimulation of anti-CD3/CD28 antibody orcecal bacterial lysate. Finallyisolated Lung CD 4’ TEM cells from ABX treated SCID mice or GF SCIDmice previously transferred with CD4“CD45RBhigh T cells indu. ced both colitis and lung manifestationswhen they were transferred to other SCID imice i’n SPF condi’tion.Conclusion : Here we have proved thatcolitogenic CD4“ TEM develiop ’Th17 mediated lung manifestationsindependent to intestinal bacterial antigenswhich may be one of the pathogenesis of extraintestinal mani-festation of IBD
