Chronic inflammation is widely implicated in gastrointestinal cancer, however, the detail mechanism by which inflammation transform normal intestinal epithelium at cellular level remains unclear due to a lack of appropriate culture system for gastrointestinal tissue. We developed a novel intestinal organoid culture system in which single Lgr5+ intestinal stem cells can indefinitely proliferate and form stereotypic structures resembling intestinal crypts. The organoid culture system could be applied to most of digestive tissue, such as stomach, colon, liver and pancreas from multiple species including human. We succeeded in culture from single clinical biopsy specimen, which enable us to research diverse gastrointestinal diseases at various statuses. I would like to show applications of organoid technology for gastrointestinal “inflammation and cancer” research in this session. |
