EGF Receptor Signaling is Essential for Initiation of Pancreatic Ductal Adenocarcinoma

抄録 EGFR and K-Ras mutations are mutually exclusive in NSCLC and in CRC, suggesting that they act in the same signaling pathway. However, ablation of EGFRs in pancreatic acinar cells completely prevents development of K-Ras oncogene-driven preneoplastic PanIN lesions and PDAC tumors. EGFR expression is also essential for PanIN and PDAC development in the context of chronic pancreatitis and in the absence of p16Ink4a/p19Arf tumor suppressors. Only loss of p53 made K-Ras oncogene-driven PDAC tumors independent of EGFR, although tumor latency and survival were considerably increased in the absence of the receptors. Combined inhibition of PI3K and Stat3 signaling pathways effectively prevents proliferation of explants derived from p53 and EGFR-deficient PDAC tumors. Thus, suggesting that loss of p53 may activate these pathways in an EGFR independent manner. These findings suggest that effective treatment of PDAC tumors may require concomitant inhibition of at least four different signaling pathways.
