抄録 The efficient and safe in vivo gene delivery to the hepatocytes is one of great milestones for gene and cell therapy treating various genetic and acquired disorders. For this purpose, we have focused on the hydrodynamic delivery that has been established as a simple, efficient, and reproducible method for delivery of DNA, siRNA, proteins, small compounds, viral vectors, and even cells in rodents. Efforts have been made to develop a clinically applicable procedure for hydrodynamic delivery to the liver in the past 10 years, which demonstrated the feasibility of applying the procedure to humans. This talk will focus on our recent work toward establishing an image-guided, computer-controlled hydrodynamic gene and cell delivery in various species including baboons. The successful application of hydrodynamic delivery in clinic will promise the safe and effective gene and cell therapy through the liver.
