Mutations of each subunit of mitochondrial complex II lead to tumourigenesis. The mechanism of this process is not clear, yet understanding of it is of importance for innovative therapies for SDH-deficient tumours. Recent studies suggest that SDH-deficient gastrointestinal stromal tumours (GISTs) accumulate succinate, leading to genome-wide DNA methylation alterations which then inhibit differentiation of the cells. Here, we have investigated whether DNA methylation is involved in cellular proliferation triggered by succinate accumulation. In DLD-1 cells, growth stimulatory effect induced by succinate under hypoxic condition was reversed by decitabine, a hypomethylating agent, suggesting the possibility that succinate promotes cellular growth through DNA methylation. |
