A retrospective analysis comparing the treatment outcomes of gemcitabine and S-1 combination therapy to gemcitabine and cisplatin combination therapy in patients with advanced biliary tract cancer

抄録 Background:Gemcitabine and cisplatin(GC)combination therapy became the standard of care for the treatment of advanced biliary cancer. In Japan, gemcitabine and S-1(GS)combination therapy also showed good tumor efficacy in phase II setting. We conducted a retrospective analysis to compare the treatment outcomes of these two treatments.
Patient and methods:Advanced biliary tract cancer patients who were treated with GS combination therapy or GC combination therapy were enrolled in this analysis. Treatment outcomes were compared between these two groups.
Results:One hundred twenty-five patients were treated with GS combination therapy and fifty-eight patients were treated with GC combination therapy. Patient characteristics were well balanced between these two groups. Response rate of GS combination therapy was significantly higher than that of GC combination therapy(28.8% vs 10.3%;p<0.01). Median time-to-progressions of GS and GC combination therapy were 5.6 months and 7.6 months, respectively(p=0.79). Median overall survival of GS and GC combination therapy were 12.4 months and 9.2 months, respectively(p=0.045).
Conclusion:GS combination therapy showed better tumor shrinkage than GC combination therapy. GS combination therapy also showed longer survival than GC combination therapy. We are now conducting a phase II study of gemcitabine, S-1, and leucovorin(GSL)combination therapy for advanced biliary cancer.
